This was the example I would have posted.  For me the Svengoolie bit is the most glorious moment.

My favourite of these is Anne Hathaway on The Daily Show, talking very seriously about her new film to John Stewart and setting it up with '... and then he gets hit by a car and he's in a coma.' There's a pause for slightly too long, then they both start to lose it at the sheer crapness of what she's describing, followed by a heroic but doomed attempt to keep going which descends into actual hysteria.

The only version I can find on YouTube is annoyingly chopped up to make a little feature, but if you jump to twenty seconds, you get most of it.

David Letterman sends Larry "Bud" Melman, an odd, older gentleman who became a bit of a mascot for the show, to New York's Port Authority bus terminal to welcome people to the city in his own unique way. The whole thing is a fun experiment in strangeness, but Dave really starts to lose it at the 3:30 mark with Larry's poor mic technique.

Can't find it on YT or owt, but does anyone recall Paul Daniels on Comic Relief, where he did a thing with some audience members or summit... at the end of his bit, which was a bit of a damp squib, Griff Rhys Jones made a quip about sending in money if you wanted to see the rest of the trick. Remember Lenny Henry absolutely losing it!
